演講摘要:

Specification of germ cells, the unique cell lineage that can deliver genetic information from generation to generation, usually takes place during embryogenesis. Like the fruit fly Drosophila melanogaster, germline specification in the pea aphid Acyrthosiphon pisum depends on maternally inherited germline determinants that are restricted to the germ plasm. ApVas1, an orthologue of the Drosophila ATP-dependent RNA helicase protein Vasa (DmVas) in the pea aphid, is one of the most conserved germline makers being identified in the germ plasm as well as the embryonic germ cells in this creature. This is the first case about the existence of a maternal germ plasm being identified at molecular basis in the hemimetabolous insects. Unlike Drosophila, where localization of Vas to the germ plasm relies on its anchorage to the Oskar (Osk) protein, germ-plasm localization of ApVas1 may be Osk-independent because the pea aphid does not have an osk homologue. We ectopically expressed various lengths of DmVas, ApVas1, and several other insect Vas proteins in Drosophila, finding that the amino acid residue glutamine (Gln) 527 within the HELICc domain of DmVas was critical for the Osk-dependent localization. Nonetheless, a corresponding residue of Gln527 was not identified in ApVas1. It thus suggests that assembly of the germ plasm in aphids depends on a non-osk molecular anchor. This presentation, I expect, can shed light on the evolution and development of germline specification in animals.
