演講摘要

 

Metastasis is the leading cause of cancer deaths. Understanding the metastatic mechanism is critical to battle this formidable disease. Here, we show paraspeckle component 1 (PSPC1) identified in an aberrant 13q12.11 locus is upregulated and associated with poor survival in cancers. PSPC1 upregulation augments tumorigenesis, epithelial-to-mesenchymal transition (EMT), stemness and metastasis in cells and mice cancer models. PSPC1 is the activator for core transcription factors of EMT, stemness and proliferation. PSPC1 interacts with Smad2/3 and dictates its binding preference from tumor suppressive to pro-metastatic targets. Together, PSPC1 is required for TGFβ1-mediated pro-metastatic signaling and a contextual determinant of pro-metastatic switch to promote metastasis.
